The arms of Mespelbrunn were granted on January 24, 1986.

The arms show the former gate of the castle of the Echter von Mespelbrunn family, as well as the arms of the family. The family originated from the village, but were of great importance in the larger region. The Julius Echter, Prince-Bishop of Würzburg, founded in Würzburg the university and hospital. The family became extinct in the late 17th century and most members were buried in the church in Hessenthal. The hasel branch in the gate is a canting symbol for Hessenthal, which is derived from Haselthal (hasel-valley). The silver and red colours are those of the State of [[Mainz (State)|Mainz]], to which the region historically belonged.
